FLIM - Fluorescence Lifetime Imaging Microscopy
 
Whole-field imaging system based on Intensified CCD Technology

LaVision BioTec offers the first FLIM system with 3D spatial and temporal resolution. It is based on the intensified PicoStar CCD camera and the advanced ImSpector software package. The system can be used for time domain (down to 50 ps) as well as for frequency domain (up to 800 MHz) measurements. The FLIM system can be combined with different kinds of laser sources, microscopes, spectrographs and LaVision BioTec's outstanding TriM Scope.

Setup

A pulsed laser excites fluorophores in the sample. An optical trigger in combination with a delay generator opens the gate of the intensified PicoStar CCD camera which measures fluorescence signal in dependance of the delay and gate time.
Gate Shift Time Domain

The acquisition of images at successive delays generates a set of images with different intensity allocations which depend n the lifetime of the fluorophores. The set of data will be processed by LaVision BioTec's ImSpector software package. Advanced software algorithms compute single as well as multi-exponential lifetime images in real time. The system also provides the functionality to work frequency domain to capture fluorescence lifetime images.
2D/3D Capabilities

2D FLIM systems work with various fs puls laserrs and wide field illumination of the sample. Whereas sectioning (3D) FLIM systems work in combination with LaVision BioTec's innovative TriM Scope technology which provides diffraction-limited 3D resolution of the fluorophores.
